Accessories Overview

On-Line Multi-Angle Light Scattering (MALS)

  • WyattQELS The WyattQELS dynamic light scattering (DLS) module integrates inside the DAWN or miniDAWN to provide simultaneous DLS measurements in the same scattering volume viewed by the MALS detectors. This integration eliminates artifacts due to band broadening or temporal displacement between separate detectors.

    In a DAWN, the optical fiber replaces any one MALS detection angle. The miniDAWN provides a separate fiber mount at 90° scattering angle.

     

  • There's more than one way to add online dynamic light scattering to a MALS detector. An alternative to the WyattQELS embedded DLS module is coupling the detection module of a DynaPro NanoStar or Mobius to a MALS flow cell via optical fiber.

    The DLS Compatibility Kit includes the optical fiber and other items needed to make the connection so the DLS batch instruments can do double duty for online DLS.

  • COMET The COMET ultrasonic transducer cleans MALS flow cells automatically by agitating the fluid and loosening particles from the glass surfaces.

    The COMET integrates inside the chassis of the MALS detector and is either controlled manually from the instrument's front panel, or automatically via an ASTRA method.

  • SEC Columns Wyatt offers silica-based SEC Columns specifically designed and tested for optimal SEC-MALS measurements of proteins and peptides.

  • WISH Injection Module The Wyatt Injection System, High-Pressure (WISH) is ideal for manual injections onto an SEC column as it triggers an ASTRA acquisition. Also useful with an Optilab dRI detector for the determination of dn/dc in flowing "batch" mode.

  • The MicroBatch Kit injects unfractionated samples directly into the flow cell of a MALS or RI detector, with the help of a syringe pump or strong thumb. It is often used in dn/dc measurements or to produce Zimm plots.

  • microCUVETTE

    Batch mode (unfractionated) measurements with the 10 µL quartz microCUVETTE are used to obtain average solution molar masses via 90° static light scattering in a MALS detector. This accessory is appropriate when molecular radii are below 10 nm and microbatch/flow cell use is not recommended, due to either the larger sample volume required or the potential for fouling.

    Scintillation vial

    Disposable scintillation vials are useful when microbatch/flow cell use is not recommended, relatively large sample volumes and high sample concentrations are available, fractionation is unnecessary or impractical, and 90° detection will not suffice. They are especially beneficial when there is a concern of samples fouling, precipitating or gelling in a flow cell or expensive quartz cuvette. The Scintillation Vial Kit (a.k.a. Batch Conversion Kit) lets you easily replace the flow cell in a DAWN MALS detector with a 20 mL scintillation vial.

    DLS

    Installing a WyattQELS module in the MALS detector adds DLS capabilities for determining average nanoparticle size (hydrodynamic radius) and even size distributions in batch mode, using the microCUVETTE or scintillation vial.

  • Orbit The Orbit helps conserve mobile phase by programmatically directing the solvent to either a waste bottle or back to the solvent reservoir.

    In a CG-MALS system, the Orbit is used to automatically select wash solvents in the post-experiment cleaning cycle.

  • Inline filters are critical to ensuring particle-free mobile phase and optimal light scattering measurements in SEC-MALS and FFF-MALS. Wyatt offers two type of inline filters for chromatography:

    • Aqueous: PEEK filter housing and titanium frit accomodate 12.5 mm (1/2") diameter filter membranes for use with aqueous solvents. Typical filter membranes are hydrophyllic PVDF.

    • Organic: Stainless steel filter housing and stainless steel filter support screen accomodate 25 mm (1") diameter filter membranes for use with organic solvents. Typical filter membranes are hydrophobic PVDF (useful for most organic solvents but not acetone, DMAc, DMF or DMSO) or PTFE (useful for most organic solvents but not benzene, chloroform, methylene chloride, toluene or xylene)

    Wyatt's inline filters are designed to be placed between the HPLC pump and the injector valve, and are not intended to filter the sample after separation. Post-column filtration can result in loss of sample components, loss of chromatographic resolution and column damage due to backpressure from clogged filters.

    These filters are not intended for use with UHPLC. UHPLC mobile phase should always be prepared to the highest standards of cleanliness in order to prevent clogging columns.

 

Dynamic Light Scattering (DLS) and Electrophoretic Light Scattering (ELS)

  • microCUVETTE Several specially-designed, high-quality quartz microcuvettes are available for batch measurements of static and dynamic light scattering. These provider higher quality data than disposable cuvettes and are necessary when samples are in non-polar solvents.

    • The MALS microCUVETTE allows static and dynamic light scattering measurements to be made simultaneously in a MALS detector, requiring only 10 µL of solution. Requires a Batch Conversion Kit.

    • The DynaPro NanoStar may be used with a unique 1 µL quartz cuvette that incorporates a solvent moat in order to prevent solvent evaporation from the sample during long measurements or in the course of a temperature ramp.

    • Both the NanoStar and Möbiuζ may utilize a 45 µL quartz cuvette, which is more convenient than the 1 µL cuvette when sufficient sample is available. An adaptor is required in order to utilize this in the Möbiuζ.

  • Disposable Cuvettes We offer several disposable cuvettes for use with the DynaPro NanoStar and Möbiuζ:

  • NanoFilter Kit The NanoFilter Kit makes it easy to filter, recover and refilter precious sample. This is especially useful when your total sample volumes are below 300 µL.

  • Atlas The Atlas pressurizes the Möbiuζ cell to allow mobility measurements of biomolecular samples, including proteins and vesicles, in high-salt buffers.

  • Mobius Dip Cell An alternative to the high-pressure flow cell, the Möbiuζ Dip Cell facilitates rapid manual measurements while requiring only 65 µL of solution under typical conditions.

 

Field-Flow Fractionation (FFF)

  • Eclipse Channels Eclipse Channels offer a wide selection of separation channels and membranes to accommodate a variety of analytical and even semi-preparative tasks:

    • Short and Long Flat AF4 Channels for optimal tradeoff between sample load, sensitivity and resolution
    • Fritless AF4 Channel to eliminate the focusing step, with concomitant high sample concentration and potential aggregation or fouling. Eclipse AF4 only.
    • Semi-Prep AF4 Channel for separating and collecting milligram quantities
    • Hollow-Fiber HF5 Cartridges for maximum convenience, highest sensitivity and lowest sample load (in the nanogram range for proteins). HF5 cartridges are usually disposed of after ~ 100 injections; replacement is fast and easy. Eclipse DualTec only.

    With flat channels you may select regenerated cellulose or poly ether sulfone (PES) membranes of varying size cutoff values. HF5 cartridges are only made of PES with 10 kDa cutoff.

  • Solaris The Orbit helps conserve mobile phase by programmatically directing the solvent to either a waste bottle or back to the solvent reservoir.

  • Solaris Solaris prevents growth of bacteria, fungi or algae in solvents with high efficiency and protects your system from down-time due to contamination.

  • Thermos The Thermos is a temperature-controlled chamber for SEC/GPC columns and FFF channels.

  • SysTrack The SysTrack module offers convenient shelving for holding multiple detectors in your experimental setup.

 

Miscellaneous

  • RI Calibration Kit Pre-made RI Calibration Standards are available to speed up RI calibration. Can be used to calibrate any Optilab or third-party differential RI instrument.

  • The MicroBatch Kit is used to inject unfractionated samples directly into the flow cell of a MALS or RI detector. It is often used in dn/dc measurements or to produce Zimm plots.

  • Solaris Solaris prevents growth of bacteria, fungi or algae in solvents with high efficiency and protects your system from down-time due to contamination.
