Light Scattering for the in-depth Characterization of
Nanoparticles and Viral Vectors

In this upcoming Gene Therapy seminar and workshop we will discuss and introduce Light Scattering as a versatile and powerful method to assess critical quality attributes (CQAs) of viral vectors (AAVs) and mRNA-based drugs such as lipid nanoparticles (LNPs).

Learn how Multi-Angle Light Scattering (MALS) and Dynamic Light Scattering (DLS) can be efficiently incorporated into your nanoparticle workflow to address identity, quantity, payload and encapsulation efficiency in addition to charge and zeta potential (PALS).

AAVs: Determine important CQAs in a single run by SEC-MALS. These quality attributes include Cp (total titer), Vg, E/F, and percentage of small aggregates. We will share case studies on using SEC-MALS as a platform method and/or in QA/AC. Additionally, AAV solutions using DLS (for screening size, particle concentration, and stability), FFF-MALS (for quantifying all AAV aggregates), and in-line monitoring (for process development) tools will be presented.

LNPs: We will discuss using SEC/FFF-MALS for multi-attribute quantification (MAQ) in one system and one single run. These quality attributes include high resolution size distribution, size-based payload distribution, and encapsulation efficiency. Tools for screening size, particle concentration, and stability with DLS, measuring zeta potential and charge with PALS, and in-line monitoring for process development (Real-Time MALS) will be presented as well.

Live software demonstration: We will demonstrate the analysis workflow in our software (ASTRA) and discuss the analysis procedure. The example data will include the analysis of an AAV and an LNP sample and what to look out for and how to set-up the experiment.

Attendees will get a refresher on Light Scattering theory with required instrumentation and explore in detail how Light Scattering can be an invaluable analytical tool for Gene Therapy and related applications.
