mRNA has risen to fame as a new and effective therapeutic modality through its extraordinary success as a COVID-19 vaccine. However, these mRNA-based vaccines would not exist without formulation by lipid nanoparticles (LNPs). The lipid shell protects mRNA from nuclease degradation and endosomal digestion when it enters the cell. As a result, LNP is becoming the preferred delivery vehicle for different types of nucleic acid (NA)-based medicines as well as other drug substances. To ensure the safety and efficacy of LNP formulated gene therapeutics, it is essential to implement robust and reliable characterization and quantitation analytics throughout the development and manufacturing process. LNP Workshop: Industrial case studies and introduction to quantifying LNP-NA CQAs by light scattering

In this workshop, we will discuss the SEC/FFF-MALS method for measuring multiple CQAs of LNP samples in a single run. These CQAs include high resolution size distribution, size-based payload distribution, and encapsulation efficiency. Strategies for using SEC/FFF-MALS in QC as a release assay will also be shared in detail. Tools for screening size, particle concentration, and stability with DLS, measuring zeta potential and charge with PALS, and in-line monitoring for process development will be presented as well.

The workshop is open to anyone who is interested in learning more about light scattering tools for LNP analysis. Admission is free, and complimentary continental breakfast is included.

LNP Advanced Method Training: How to quantify LNP-NA CQAs by light scattering
11:00 AM - Open to all

We will provide step-by-step training on how to use SEC/FFF-MALS and Wyatt’s LNP Analysis to quantify LNP-NA CQAs using the LNP Guidance Manual as reference. We will also provide guidance on best practices on using DLS and PALS for this application. At the end of the meeting, we will offer data review opportunities to those who do not mind staying late. Admission is free, and a complimentary lunch will be provided.
