AAV Workshop: An introduction to the characterization of AAV by light scattering

Adeno-associated virus (AAV) is an attractive delivery vehicle in gene therapy attributed to its mild immune response and ability to deliver the therapeutic genome into a wide range of host cells. To ensure the safety and efficacy of the viral vector-based drug product, it is essential to implement robust and reliable characterization tools throughout the production and manufacturing process. In this presentation, we will discuss a size exclusion chromatography (SEC) method coupled with UV, multi-angle light scattering (MALS), and differential refractive index (dRI) detectors to measure the following three important AAV quality attributes (QAs): 1) total number of viral capsid particles; 2) relative capsid content (e.g., ratio of empty and full capsids); and 3) percentage of monomer or aggregates. Additionally, orthogonal methods using dynamic light scattering (DLS) and field flow fractionation (FFF) will be compared.

AAV User Meeting: Secrets to the successful quantification of AAV by light scattering

We will provide step-by-step training on how to use SEC-UV-MALS-dRI, protein conjugate analysis in ASTRA software, and additional calculations to measure AAV quality attributes (QAs). The QAs include: 1) total number of viral capsid particles; 2) relative capsid content (e.g., ratio of empty and full capsids); and 3) percentage of monomer or aggregates. We will also be available to answer any of your questions regarding DLS and FFF applications and analysis.
